Check out Spiv's vblank demo, I think that uses the timer interrupt to simulate the vblank (or hblank, can't remember of hand).
1) Disable interrupts, call a SWI routine with a pointer to your interrupt routine.
2) As many as there are vectors. I think there's 31.
3) None I believe as the gp32 contains no OS as such. Oh, apart from the SWI of course. USB interrupt shouldn't be modified either really unless you intend replacing it like in GpDrive (which is another good example of an interrupt).
